Antithrombotic, Antiaggregant and Anticoagulant Effect of Methanolic Fraction of Calamintha ofcinalis: In vitro and ex vivo Experiments

Abstract
The objectve was to study the effect of Calamintha ofcinalis methanolic fracton in the preventon of thrombosis and to explore its effect on primary and secondary hemostasis. The antthrombotc actvity was evaluated by using the pulmonary thromboembolism experimental model. Platelet aggregaton, tail bleeding tme and coagulaton assays were explored to elucidate the antthrombotc mechanism. For aggregaton, washed platelets were incubated in presence of Calamintha ofcinalis methanolic fracton and then actvated by different agonists (thrombin, ADP, collagen). The bleeding tme was determined to measure the tme between the tail incision and the moment the bleeding stops. The coagulaton was explored by measuring prothrombin tme, actvated partal thromboplastn tme, thrombin tme and plasmatc fbrinogen concentraton. For ex vivo study, the animals were treated by the Calamintha ofcinalis methanolic fracton (250 mg/kg/day) for 4 weeks. Then, platelet aggregaton, tail bleeding tme and coagulaton parameters was studied. The results indicate that C. ofcinalis methanol fracton seems to exert in vivo an antthrombotc acton (40% of protecton). The in vitro investgatons also show that this fracton signifcantly inhibits aggregaton, increases bleeding tme, coagulaton tmes and reduces fbrinogen concentraton. The chronic treatment of animals confrms the precedent data. This study demonstrates an antthrombotc effect of C. ofcinalis. The observed antaggregant and antcoagulant effects could explain this actvity
