Advances in understanding the mechanism of cancer from the perspective of ribosomopathies

Abstract

Ribosomes are one of the widespread and numerous organelles, whose abnormal biogenesis and function can cause ribosomopathies, which are characterized by high cancer predisposition. Studies have shown that the pathophysiological mechanism of ribosomopathies is also the carcinogenic mechanism of many sporadic tumors, such as p53 signaling pathway activation and protein translation disturbances. In cancer cells, ribosome biosynthesis significantly changes, whose heterogeneity is closely associated with the occurrence, development and metastasis of cancer. In this paper, we summarize the characteristics and pathophysiological mechanism of ribosomopathies, and understand the pathogenesis of sporadic cancer from the perspective of ribosomopathies, which is expected to provide an important theoretical basis for ribosome-targeted cancer therapy.
