An efficient protocol for genomic DNA extraction from Capparis Spinosa L

Abstract

This research explained an efficient method of DNA extraction in Capparis Spinosa L. The Quality of DNA were studied with CTAB method with and without liquid nitrogen. This protocol was modified by published protocol. DNA visually checked in compare with lambda band in 0.7% agarose gel and products of PCR.A method is described for Isolation of high quality DNA from 5 populations Capparis Spinosa in South of Iran using CTAB method without liquid nitrogen. These plants typically contain high levels of mucilages, polyphenols, secondary metabolites, antioxidant, complex polysaccharide compounds, thus extraction of DNA by common methods is a hard task. This method change into previous published protocols which is more efficient, proper and rapid for DNA extraction to improve the quality of extracted DNA from plants. Here, the original protocol is modified. The efficiency of these changes result in high-quality DNA ready to use in a variety of molecular biology analysis methods, such as PCR, sequencing and cloning which yield into good results. The effectiveness of this method is proved by the amplification and sequencing of PCR products .This versatility is not usually found in DNA extraction protocols. In addition, this method is adaptable to standard laboratories, and most importantly it is quicker, safer and more cost-effective. The method of extraction can also be used for Capparidaceae family and the same plants. The best quality was seen in extracted DNAs from published modified method that shown in Fig 3. After dilution it used in PCR reaction (Fig.4) that resulted in purify bands of PCR..
