Construction and partial sequencing from a subtractive library in cut Calcutta 4 ’(Musa AA) in early stage of infection with Mycosphaerella fijiensis Morelet
Journal Title: Biotecnología Vegetal - Year 2006, Vol 6, Issue 4
Abstract
The study of genes involved in plant defense response against pathogen attack, is one of most important steps leading to the elucidation of disease resistance molecular mechanisms. The generation of subtracted deoxyribonucleic acid libraries (cDNA), by means of suppression subtractive hybridization technique (SSH), has been used for this purpose. A subtractive hybridization was made between a cDNA population obtained from ‘Calcutta 4’ inoculated leaves with M. fijiensis (CCIBP-Pf83) and a mixture of cDNA from ‘Calcutta 4’ non inoculated leaves and mycelium. Leaves samples were taken at 6, 10 and 12 days after inoculation. The subtracted library was obtained by cloning and transformation of subtracted products and as a result, 600 recombinants clones were obtained. Sequence analysis of sixty nine clones, revealed redundancy of the expressed sequence tags and most of them showed no homology with reported sequences at databases and only 13 % had a high homology with metalothioneins. The results constitute a step in advance in the molecular study of Musa-Mycosphaerella fijiensis interaction.

Effect of water stress induced by PEG 6000 on in vitro germination of Phaseolus vulgaris L. cv. ‘ICA Pijao’ seeds
The common bean (Phaseolus vulgaris L.) production has been very important in Cuba, however, several factors affect the yield of this crop and not all cultivars of beans respond the same way to the stressful conditions...
Growth, regeneration and radiosensitivity of sugarcane callus (Saccharum spp. Hybrid var. "SP 70-1284") treated with
Calluses in growth of sugar cane, variety SP 70-1284, were radiated with a source of 60Co. The studied doses were between 10 and 80 Gy. The affectation of the Gamma radiation source 60Co in the growth of the callus and...
Effect of immersion frequency on shoot multiplication of Bambusa vulgaris Schrader ex Wendland in RITA ® TIS
Bamboos are unique in bringing together a wide range of environmental services. The present work was conducted to determine the effect of immersion frequency of shoot multiplication of B. vulgaris grown in SIT. Three Im...
Analysis of the mineral nutrients content during the formation and maturation of somatic embryos of ‘FHIA-21’ (Musa AAAB)
Despite the advances reached using somatic embryogenesis in Musa spp., there are not studies that state the requirements of mineral nutrient during its development. This investigation aimed to determine the mineral nut...
Somatic embryo formation in Glycine max variety Incasoy-27
