Down-regulation of thymic stromal lymphopoietin by curcumin.
Journal Title: Pharmacological Reports - Year 2013, Vol 65, Issue 2
Abstract
Background: Thymic stromal lymphopoietin (TSLP) is a cytokine implicated in the pathogenesis of allergic diseases such as asthma, atopic dermatitis and allergic rhinitis. Curcumin has various effects such as antidepressant, antioxidant, antihyperglycemic, antitumor and anti-inflammatory. However, the effect of curcumin on the production of TSLP has not been clarified. Thus, we investigated how curcumin inhibits the expression and production of TSLP in the human mast cell line, HMC-1 cells. Methods: We used enzyme-linked immunosorbent assay, reverse transcription-polymerase chain reaction, luciferase assay, and caspase-1 assay to investigate the effects of curcumin. Results: The results show that curcumin inhibited the production and mRNA expression of TSLP in HMC-1 cells: the maximal inhibition rate of TSLP production by curcumin (50 μM) was 59.16 ± 4.20%. In addition, curcumin suppressed the nuclear factor-κB luciferase activity induced by phorbol myristate acetate plus A23187. In the activated HMC-1 cells, caspase-1 activity was increased, whereas caspase-1 activity was decreased by pretreatment with curcumin. Conclusion: These results suggest that curcumin can be used to treat inflammatory and atopic diseases through the suppression of TSLP.

Monocyte-suppressing effect of high-dose metformin in fenofibrate-treated patients with impaired glucose tolerance.
Background: Fibrates were found to reduce cytokine release and low-grade inflammation in patients with impaired glucose tolerance. The aim of this study was to investigate whether these effects of fibrates may be potenti...
Hypertriglyceridemia but not hypercholesterolemia induces endothelial dysfunction in the rat.
In humans, hypercholesterolemia and hypertriglyceridemia induce endothelial dysfunction and therefore lead to atherosclerosis. In contrast, rats are resistant to atherosclerosis. Here we analyze whether rats respond to h...
Systemic perfluorohexane attenuates lung injury induced by lipopolysaccharide in rats: the role of heme oxygenase-1.
Clinical trials with partial liquid ventilation demonstrate improvement in oxygenation, as well as some adverse side effects linked to the application of liquid perfluorocarbons (PFCs) during liquid ventilation. Thus, we...
Potential neurotoxic effect of ethylene glycol ethers mixtures.
Background: Ethylene glycol ethers (EGEs) are widely used as mixtures in various industrial processes and in many household products. 2-Methoxyethanol and 2-ethoxyethanol primarily exert gonadotoxic effect, while 2-butox...
Effects of some convulsant agents on the protective activity of topiramate against maximal electroshock-induced seizures in mice.
The anticonvulsant activity of topiramate combined with some convulsant agents (bicuculline--BIC, N-methyl-D-aspartate--NMDA, and kainic acid--KA), given at subconvulsive doses, was evaluated in the maximal electroshock...
