Effect of carborundum damage on regeneration and expression a-glucuronidase transient in different explants of Phaseolus vulgaris var. CIAP 7247

Abstract
Agrobacterium tumefaciens-mediated genetic transformation is difficult in beans. Efficiency can be improved preparing explants for the infection. The aim of this research was to determine the effect of treatment with carborundum to cotyledon nodes and organogenic callus of Phaseolus vulgaris var. CIAP 7247 in the regeneration and transient expression of â-glucuronidase. A stem segment of 5-6 mm length, containing the cotyledon node, was taken three days after mature seed germination. Fragments of 4-5 mm diameter on the seventh day after the third subculture were used in the case of organogenic callus. Five treatments at different times for the application of carborundum were studied (0, 0.5, 1.0, 1.5 and 2.0 minutes). Results showed that the application of a carborundum solution (0.5g/30ml of sterile water) in agitation, caused damage on both types of explants used. These results concluded that the application of carborundum for 1.5 minutes achieved at least regeneration of multiple buds and an average 9.68 of blue spots. However, there was an increase GUS positive spot in organogenic callus with a treatment for 1.0 minute, without affecting subsequent plant regeneration of transformed tissues in this variety of beans.
