EFFECT OF OREXIN DRUG AND SOME MINERALS ON RATS EXPOSED TO OBESITY AND ALZHEIMER DISEASE

Abstract

The affect of administration of some minerals on rats exposed to obesity and Alzheimer disease was studied. Thirty rats of Sprague Dawley (weighting 200±10 g) were divided into sex groups (5 rats). One group considered as normal control group. Twenty five rats were fed on the high fat diet and administered aluminum chloride for 6 weeks to induce obesity and Alzheimer The obese with Alzheimer's rats were classified into positive control, orexin (20 mg/kg b.w / i.m), iron (100 mg/kg b.w), calcium (6 g/kg b.w) and zinc (18 mg/kg b.w) by stomach tube groups. The results revealed that there was a significant decrease in body weight gain, weight gain percent and FER among all rats group which treated with orexin, calcium, iron and zinc in comparing with non- treated obese male and Alzheimer's rats positive control group. Moreover, The treatment rat groups showed significant decrease in serum cholesterol (CHO), triglyceride (TG), low density lipoprotein cholesterol (LDLc), very low density lipoprotein cholesterol (VLDLc) and total lipids (T. Lipids) and also decrease in atherogenic indexes but showed significant increase in serum high density lipoprotein cholesterol (HDLc), iron and zinc in comparing with non-treated positive rat group. However, These rat groups showed a significant increase in brain catalase (CAT), glutathione-S- transferase (GST) and superoxide dismutase (SOD) but a significant decrease in brain levels of malondialdehyde (MDA) level and B-amyloid compared to positive control group. These biochemical changes were accompanied with histopathological alterations in brain tissue. It is concluded that the treatment with minerals could ameliorate most of the evaluated biochemical parameters and improve the induced degenerative histopathological changes.
