Evaluation of the cell-free coelomic fluid of the earthworm Eudrilus euginiae to induce apoptosis in SiHa cell line

Abstract
The cell-free coelomic fluid (CF) was collected from the earthworm Eudrilus euginiae and tested for toxicity at 20, 40, 60, 80 and 100 µl.ml -1 concentrations by MTT assay in SiHa cells invitro. Percentage of inhibition for SiHa cells invitro by CF recorded upto78.52% at 100 µl.ml -1 concentration in 48 hours. AO/EB and Hoechst 33258 staining showed signs of necrosis at 100 µl.ml -1 but morphological changes of typical apoptosis was observed at 20 and 40 µl.ml -1 concentrations. Apoptotic rate increased from 38.58 % to 59.21 %, when repeated at 40 and 50 µl.ml -1 concentrations of CF in 48 hours. Similar increase of apoptotic rate from 68.91% to 79.32% was observed with the increase in the incubation period from 48 h to 72 h suggesting that inducement of apoptosis in SiHa cells invitro by the CF was a function of dose and time period. SDS-PAGE analysis of the CF revealed the presence of a 42 KDa protein – identified earlier to be a tunorilytic factor analogous to the vertebrate cytokine Tumor Necrosis Factor (TNF). Tumorilytic activity of the CF of Eudrilus eugeniae has not been recorded earlier and therefore suggests further investigations in this animal model.
