Exploration into the mechanism of Huatan Tongluo decoction in the treatment of cerebral stroke based on network pharmacology

Abstract

[Objective] To analyze the mechanism of Huatan Tongluo decoction in the intervention of cerebral stroke by network pharmacology. [Methods] The effective components and potential targets of Huatan Tongluo decoction were screened through TCM System Pharmacology Database and Analysis Platform (TCMSP) and BATMAN-TCM database, and the cerebral stroke related targets were screened using human gene database (GeneCards) and online human Mendelian genetic database (OMIM). The common target of Huatan Tongluo decoction and cerebral stroke were obtained based on the online software Venny analysis, and Cytoscape software was used to construct the visual network diagram of Huatan Tongluo decoction-cerebral stroke common target. The protein interaction (PPI) network diagram was constructed by using STRING database, and the PPI network topology analysis was carried out on the common target. Screen out the core target. Rstudio software was used to visually construct the top 10 GO analysis and the top 20 KEGG path enrichment analysis of the common target. Finally, the KEGG pathway relationship network diagram was constructed by Cytoscape software. [Results] A total of 109 active components and 294 corresponding targets of Huatan Tongluo decoction were selected from the database, 158 targets were common with stroke, among which 23 were core targets. Sitosterol, stigmasterol, baicalein and naringin were the main effective components of Huatan Tongluo decoction. Core targets include TP53, CCL2, HIF1A, ESR1, etc. GO enrichment analysis mainly involved biological processes such as response to xenobiotic stimulus, response to decreased oxygen levels, and response to oxygen levels. KEGG pathway enrichment analysis showed that lipid and atherosclerosis, AGE-RAGE signaling pathway, cAMP signaling pathway, apoptosis may be the mechanism of Huatan Tongluo decoction in the intervention of cerebral stroke. [Conclusion] Huatan Tongluo decoction may act on different targets through various components, and play a therapeutic role in stroke by regulating lipid, inhibiting apoptosis, reducing inflammation, protecting blood vessels and other pathways.
