Expression of cidea, cyr61, azgp1, ask1, and hlf genes in subcutaneous adipose tissue of obese men with and without glucose tolerance

Abstract
We studied the expression of CIDEA, CYR61, AZGP1, ASK1, and HLF genes in subcutaneous adipose tissue of two groups of obese men: with normal and impaired glucose tolerance as compared to control group (lean subjects). It was shown that the expression level of CIDEA, AZGP1, ASK1, and HLF genes is decreased in subcutaneous adipose tissue of obese men with normal glucose tolerance as compared to control group. At the same time, the expression of CYR61 gene is increased in subcutaneous adipose tissue at these experimental conditions. Magnitude of obesity-mediated changes in expression levels of these genes was gene specific and more robust in the case of AZGP1 gene. In the group of obese men with glucose intolerance the level of CYR61 gene expression in subcutaneous adipose tissue is also increased as compared to control group, but was significantly lower than in obese patients with normal glucose tolerance. At the same time, the expression of ASK1 and CIDEA genes is increased in subcutaneous adipose tissue in the group of obese men with glucose intolerance versus obesity group without impaired glucose tolerance. Results of this study provide evidence that the expression of CIDEA, CYR61, AZGP1, ASK1, and HLF genes is changed in subcutaneous adipose tissue of obese men with normal glucose tolerance and these changes possibly associated with developing of obesity and metabolic abnormalities. However, impaired glucose tolerance in obesity is associated with up-regulation of ASK1 and CIDEA genes and down-regulation of CYR61 gene expressions.
