Formulation and evaluation of biodegradable Ondansetron hydrochloride implant

Abstract

Implantable drug delivery systems are used to deliver drugs for longer period of time, beneath the skin. These drug delivery systems are implanted subcutaneously and drug is supposed to be released in sustained manner for the desired time period. Subcutaneous route is selected for the sustained and prolong release of drugs, Ondansetron which is a selective antagonist of serotonin 5-HT3 receptors and also effective for attenuating the abuse of a variety of psychotropic substances, including alcohol. This route is selected because it avoids first pass effect metabolisms & other type of drug loss through different organs of the body. In the present piece of investigation an attempt has been made to design and develop biodegradable subdermal implants of Ondansetron HCl. Tablet shaped implants were prepared with the help of cross linked Chitosan. Preformulation studies for drug as well as polymer have been done using UV spectroscopy, Infrared spectroscopy and DSC. The implants were evaluated for drug content uniformity, thickness, weight variation and in vitro dissolution studies. All developed implant formulations followed Zero order kinetics of drug release and Korsmeyer Peppas kinetics indicting that drug release from implant formulations is controlled by diffusion of drug as well as erosion of polymer.
