Functionalization of vinyl cubic Ia3d mesoporous silica for immobilization of penicillin G acylase

Abstract
Vinyl cubic Ia3d mesoporous silica (V-CIMS), prepared by co-condensation method, was epoxidized by m-Chloroperbenzoic acid to epoxy group-functionalized mesoporous silica (E-CIMS). The above mesoporous silicas were characterized by X-ray diffraction (XRD), N2 sorption, thermogravimetry-differential thermal analysis (TG-DTA) and transmission electron mircoscopy (TEM), which shows that the framework of cubic Ia3d silica of mesoporous silica is not destroyed after the treatment by H2SO4 to remove the templates and the functionalization of vinyl to epoxy group. Penicillin G acylase has been successfully immobilized covalently on E-CIMS supports, and PGA/E-CIMS (molar ratio of TEVS and TEOS=1∶9) behaves the initial specific activity of 2 262 IU/g and the best operational stability of 80% of the initial specific activity after recycled for 5 times.
