In vitro establishment of Morera

Abstract

Apical buds like explantes were used which were established in the culture medium proposed by Murashige and Skoog. In the disinfection sodium hypoclorite was used to the 0.5, 1.0 and 1.5% during 10, 15 and 20 minutes and the combination of sodium hypoclorite and ethanol 70%, evaluating microbial contamination (%) and survival (%). In the establishment it evaluated the Influence of the 6-BAP and kinetin using 0.5, 1.0 and 2.0 mg.l -1 in each one of them, culture medium semisolid as liquid and the sprouted explants was evaluated (%), height of buds (cm) and survival (%). With the combination of sodium hypoclorite 1% and ethanol 70% (p<0.05) it decreased to 0.9% the microbial contamination and to improve to 98.4% the survival of explants. In the establishment better results were observed with 6-BAP compared with kinetin and the best treatment was using 0.5 mg.l -1 6-BAP the 97.4 and 98.2% of the explantes sprouted respectively in the culture medium semisolid and liquid being obtained, being statistically different (p <0.05) with the other treatments; in the same way for height of buds which was of 4.91 and 5.06 cm and, the lowest value in 2.42 and 2.8 cm were observed in the control.
