Influence of growth regulators on callus formation of Phaseolus vulgaris L cv. CIAP 7247

Abstract
The present work had as objective to achieve the formation of nodular calli in Phaseolus vulgaris L. cv. CIAP 7247 using several growth regulators in the culture medium. Cotyledons of mature seeds without coats were used as explants. The effect of different concentrations of TDZ, 2,4-D and AIA was evauated for the callus formation. In the treatment with different concentrations of TDZ, the explants showed green-yellow coloration during the first two weeks of culture. When the callus growth was evaluated the best values were obtained when 0.2 mg.l -1 TDZ was used. When the effect of 2,4-D was evaluated in the culture medium were not found significant differences in the explants number with callus formation for the studied concentrations. These were not compact, have watery consistency and yellow-whitish coloration without presenting visually characteristic of nodular calli. Nevertheless, those formed in the culture medium with AIA were less voluminous, but they presented better characteristics in their consistency. It was demonstrated that the combination of TDZ-AIA was the best for the formation of nodular calli in Phaseolus vulgaris cv. CIAP 7247.
