Tinzaparin Tedavisinin Oral Yassı Hücreli Karsinoma Çıplak Fare Modelinde BAX ve CCNB1 Gen Anlatımları Üzerindeki Etkisi

Abstract

The Effect of Tinzaparin Treatement on BAX and CCNB1 Gene Expression in an Oral Squamous Cell Carcinoma Nude Mice Model Introduction: This study aims to evaluate the effect of tinzaparin treatment on the expression of proapoptotic BAX and cell cycle G2/M phase regulator CCNB1 genes in an oral squamous cell carcinoma (OSCC) nude mice model. Materials and Methods: Total of 11 nude mice at 4–6 weeks of age, composed of three groups [tinzaparin treated (n=4), positive control (n=4), negative control (n=3)], were used to create an orthotopic OSCC mice model. Human tongue cancer cell line OSC-19 was utilized to create this in vivo model. After 21 days, the mice were sacrificed and tumors were quantified for BAX and CCNB1 expression utilizing quantitative real-time polimerase chain reaction (qRT-PCR). Livak method normalized for control samples revealed fold-changes utilizing ACTB housekeeping gene. Statistical significance was tested by non-parametric Mann-Whitney U test, where P<0.05 was considered significant. Results: Two mice in positive control and tinzaparin groups did not develop tumors. Available tumors showed decreased BAX and CCNB1 expressions with low statistical significance, when compared to the control group. Conclusion: These preliminary results indicate a deregulated cell cycle control without an effect on apoptosis. More mice should be included to significantly state the effect of tinzaparin treatment in OSCC mice model. Keywords: Oral squamous cell carcinoma, nude mice, qRT-PCR, BAX, CCNB1
