Functional analysis of miR161.1 by STTM technology in Arabidopsis

Abstract
[Objective] The miR161.1 knockdown mutant was obtained by STTM technology. The STTM161.1 mutant plants displayed significant phenotypic alterations. It lays a foundation for the functional study of miR161, and can be applied to sterile seed production of crop hybrid. [Methods] In Arabidopsis, the transgenic lines, STTM 161.1, were generated by the transformation of the constructed pFGC5941-STTM161.1 binary vector. STTM161.1 mutant plants were obtained by genotype and phenotype analysis. Transcriptome analysis was performed to identify the alterations of genome-wide gene expression. [Results] RT-qPCR analysis indicated that miR161.1 down-regulated in STTM161.1 plants, while target genes were significantly up-regulated. Compared with the wild type, STTM 161.1 plants displayed various of phenotypic alterations, including early reproductive age, enlarged leaves, partial male sterility, and seed abortion in fruit pods. The transcriptome analysis revealed that several plastid expression genes were significantly up-regulated, and miR161.1 was mainly involved in multi-ple metabolic pathways, including secondary metabolite biosynthesis, phenylpropane biosynthesis, purine metabolism, and beta-linolenic acid. The differentially expressed genes screened between STTM161.1 and the wild type include a MADS TF encoding gene associated with floral development, and a bHLH transcription factor encoding gene associated with plant floral and pollen development. [Conclusion] miR161.1 was proved to have taken part in the regulation of male sterility, developmental phase transition and seed development in Arabidopsis. 17 up-regulated and 6 down-regulated differentially expressed genes were verified to be possibly related to the functions of miR161.1.
