TABLETING PROPERTIES OF THERMALLY ACTIVATED COW BONE POWDER IN LOW DOSE FORMULATION

Abstract

 ABSTRACT Formulations of low dose drugs into a tablet dosage form require appropriate diluents in a significant quantity that may enhance the ease of handling. Activated cow bone powder was evaluated and found suitable as a diluent in the formulation of low and high dose drugs. This study was conducted in order to compare the diluent properties of dicalcium phosphate (DCP) and activated bone powder (ABP) in the formulation of low dose drugs. The mixed powders were pelletized with the specac pelletization machine, using a compaction force between 10-20 metric tonnes. The weights of 250 mg of granules were put in the die of 8.5 mm punch and were compressed using Erweka single station tableting machine. The formulated folic acid tablet was evaluated for properties such as weight uniformity, thickness, crushing strength, friability, disintegration and organoplastic. Assay of the tablet was done using spectrophotometric analysis. Of interest in this formulation is the assay which gave as much as 96% content and the weight variation which is below 2%. In all the formulations, ABP compressibility was higher than that of DCP. The disintegration times were within the acceptable time limit. The hardness of the tablets containing same formulation with ABP compared with DCP was higher in all cases. The friability values generally were lower than 1%, which is the limit accepted by pharmaceutical industries. Analysis of the granule size distribution of the pellets gave a mean granule size (MGS) of 442µm. The disintegration time and the friability values for all tablets from the single station tablet machine and the pelletizer were all within the acceptable official limits. Activated bone powder was found to be better direct compressible diluents than the commercially available dicalcium phosphate for low dose drugs such as folic acid.
