Formulation and evaluation of Diclofenac potassium transdermal patches for enhanced therapeutic efficacy

Abstract
Today 74% of the drugs are taken orally and are found not to be safe or effective as desired because they produce wide ranging fluctuation in drug concentration in blood stream. Continuous I.V infusion is recognized as a superior mode of drug administration with certain limitations like patient hospitalizing and monitoring. Recently it is evident that I.V infusion can be duplicated by using the skin as a part of drug administration to provide continuous transdermal drug infusion in systemic circulation by non-invasive method. Transdermal Drug Delivery System represents one of the most rapidly advancing area of novel drug delivery. In the present work transdermal patches of diclofenac potassium was prepared by using different ratios of polymers like PVA and PVP plasticized with glycerin and Dimethyl sulfoxide by solvent casting method with the objective to improve its therapeutic efficacy, Patient compliance, as well as to avoid its extensive first pass metabolism. The formulated TDDS patches were evaluated for various physicochemical parameters like thickness, uniformity weight, folding endurance. From the studies it was found that the formulated patches with different polymer concentration were found to be Flexible, Smooth, non-sticking and homogeneous in nature Thickness and average weight of the patches did not show any significant differences. All the 6 formulations have showed good folding endurance indicated that the patches have good flexibility. In vitro dissolution studies were carried out for 6 formulations and foundF4 showed optimum drug release and was selected as an ideal formulation for further release kinetic studies and in vivo studies.
